Gab2遺伝子のノックアウトマウス。Gab2はアダプター分子であり、様々なチロシンキナーゼによってリン酸化を受ける。リン酸化されたチロシン残基はSH2ドメインを有するPI-3キナーゼやSHP-2といったシグナル伝達分子と会合する。Gab2欠損マウスでは、マスト細胞の発生障害が観察され、アナフィラキシーに対して、抵抗性を示す。マウスではGab1とGab2のファミリー分子が報告されている。サイトカイン・増殖因子を介するシグナル伝達の解析、アレルギーに関する研究に有用。

Gab2 gene knockout mice. Exon encoding a major portion of the PH domain was replaced with a neo cassette. Gab2 gene is an adaptor molecule that plays a critical role in mediating cellular transformation. The number of mast cells is severely reduced in the stomach and less severely in the skin of homozygous mutant mice. Bone marrow-derived mast cells from the homozygous mutant mice exhibit an impaired KitL-induced ERK MAP kinase and Akt activation.
